Trey was born in Conifer, Colorado, on October 19, 1969 to Randy Parker, a geologist, and Sharon Parker, an insurance broker. He has an older sister, Shelley Parker. He met Matt Stone (co-creator of South Park (1997)) while attending the University of Colorado in Boulder, where he had a double major of music and Japanese. While at UCB he wrote, directed and starred in Cannibal! The Musical (1993) (aka "Cannibal: The Musical!") based on a true episode in Colorado's history. After graduation from UCB (rumors that he didn't due to skipping classes to work on the movie are false), he and Stone were asked by then-FoxLab executive Brian Graden to create an animated Christmas card for his friends and family. The now infamous short, titled The Spirit of Christmas (1995), led to South Park (1997).

Parker's nickname, Trey, comes from his original name, Randolph Severn Parker III. He is the third person in his family with that same name, and Trey means "Three" in some languages. On South Park (1997), he voices, among other characters, "Stan Marsh" and Stan's father, "Randy Marsh". Parker gave Stan's parents the same names as his own parents (Sharon and Randy).
Parker has played piano since the age of 12 and credits Elton John as a hero. He also sings and plays the keyboard for a band called D.V.D.A.
Has a black belt in Taekwondo.
Parker wore the green, low-cut dress popularized by singer Jennifer Lopez to the 72nd Academy Awards while Stone wore the pink dress styled by actress Gwyneth Paltrow.
His character, Eric Cartman, was rated number 16 in Comedy Central's newest show 'Mouthing Off: 51 Greatest Smartasses'.
He had written a script titled "Fuzzies", which is a children's story he was planning to direct using stop-motion-animation for the closet creatures, but the project has been put on hold. He and Matt Stone were also planning to make a movie based on "That's My Bush!".
Always hoped that Saddam Hussein would see his movie South Park: Bigger, Longer & Uncut (1999), in which he and Matt Stone made fun of him. In 2004 Parker and Stone portrayed another active dictator, Jong-Il Kim of North Korea, in their new movie Team America: World Police (2004). Two days after the premiere, they sent a copy to the North Korean government.
Nearly every song heard on South Park (1997) is written by Parker. He also composed every new song for Team America: World Police (2004), including "Freedom Isn't Free", "America F*** Yeah!", and "The End of an Act", as well as "Everyone Has AIDS", the last of which he co-wrote.

On March 5, 2019, Trey Parker filed for divorce from his wife Boogie Parker (nee Tillman) citing irreconcilable differences through the Los Angeles court. The couple stopped living together on the 28th of February, 2019. Trey has filed for joint custody of their daughter and the terms of their pre-nuptial agreement discourage her from seeking spousal support.
