Betty Norton ( Mae Murray ) and her younger brother Jimmy ( Matty Roubert ) have run away from Nifty Mendez, a white slaver. While on he run, Jimmy's leg is broken when hit by a car. The car's wealthy owner Rodney Channing ( Harry C. Browne ) insists Jimmy stay with him during his recuperation. Rodney and Betty fall in love during this time and begin to make wedding plans. Mendez discovers their plans and demands money from Betty, threatening to expose the fact that her father is in jail if she doesn't comply. The distraught girl writes Rodney a letter, telling him the truth about her father, then she runs away. Mendez is murdered by an old enemy and Rodney is finally able to locate Betty, reassuring her of his unconditional love and they begin again to make their wedding plans.

The Big Sister was produced by the Famous Players Film Company and released in September, 1916. Advertised as, Presenting the Fascinating Mae Murray in a powerful drama which thrillingly illustrates how the " other half " lives. It is now listed as unknown on the long list of lost silent films. Fans of Mae Murray and early silent films hope for a copy of this drama to resurface.
